S-genotype identification, genetic diversity and structure analysis of Italian sweet cherry germplasm
2017
In this study, 186 local sweet cherry accessions from 12 Italian regions, plus eight reference accessions, were analysed for the first time, using 13 microsatellite markers. Moreover, their S-incompatibility genotypes were identified with consensus primers for the S-RNase and SFB genes. A total of 161 unique genotypes were found; 18 groups of synonyms, along with the discovery of cases of misidentification. The average number of alleles per locus was 9.7, the mean expected heterozygosity (He) was 0.63, the mean observed heterozygosity (Ho) was 0.65 and the mean polymorphic information content (PIC) was 0.58. The structure analysis revealed the presence of six populations, which reflected in…
Algerian Olive Germplasm and Its Relationships with the Central-Western Mediterranean Varieties Contributes to Clarify Cultivated Olive Diversificati…
2021
Olive tree with its main final product, olive oil, is an important element of Mediterranean history, considered the emblematic fruit of a civilization. Despite its wide diffusion and economic and cultural importance, its evolutionary and phylogenetic history is still difficult to clarify. As part of the Mediterranean basin, Algeria was indicated as a secondary diversification center. However, genetic characterization studies from Maghreb area, are currently underrepresented. In this context, we characterized 119 endemic Algerian accessions by using 12 microsatellite markers with the main goal to evaluate the genetic diversity and population structure. In order to provide new insights about …
Parentage Atlas of Italian Grapevine Varieties as Inferred From SNP Genotyping.
2021
The Italian grape germplasm is characterized by a high level of richness in terms of varieties number, with nearly 600 wine grape varieties listed in the Italian National Register of Grapevine Varieties and with a plethora of autochthonous grapes. In the present study an extended SNP genotyping has been carried out on Italian germplasm of cultivated Vitis vinifera subsp. sativa and Vitis hybrids. Several hundred Italian varieties maintained in the repositories of scientific Institutions and about one thousand additional varieties derived from previous studies on European, Southern Italy, Magna Graecia and Georgian germplasm were considered. The large genotyping data obtained were used to ch…
Genetic diversity in a collection of Italian long storage tomato landraces as revealed by SNP markers array
2019
Tomato (Solanum lycopersicum L.) is one of the most important crops worldwide. In this study, we used 7720 genome-wide SNPs to characterize the genetic diversity within a tomato germplasm collection enriched with 64 accessions from southern Italy of the so called "da serbo" type i.e. drought-tolerant and long storage landraces. Notwithstanding the relatively small collection area, 1575 (20.4%) polymorphic SNPs, mostly on Chr11, detected considerable levels of genetic variation. Maximum parsimony analysis of genetic distance revealed four main clusters and clearly separated most "da serbo" landraces from the outgroups. One of the clusters grouped the landraces from the Mount Vesuvius area, t…

In vitro propagation of the relict laperinne’s olive (Olea europaea L. subsp. laperrinei)
2017
Olea europaea L. subsp. laperrinei (Oleaceae) is an endemic taxon of the mountainous regions of central Sahara, consisting of currently fragmented and small relict populations. The tree can propagate vegetatively or by seed, but no recent natural regeneration was observed in the Algerian massive populations, some of which are considered threatened with extinction. Sterile triploid individuals were also identified in some populations showing increasing vigour. As a result of its long persistence and despite its rarity, the Laperrine's olive is an iconic component of Saharan mountain ecosystems. The aim of this study is to develop an efficient micropropagation protocol for both diploid and tr…
Organogenesis and encapsulation of in vitro-derived propagules of Carrizo citrange [Citrus sinensis (L.) Osb. × Poncirius trifoliata (L.) Raf]
2011
Due to widespread polyembryony, Citrus rootstocks are usually propagated by open-pollinated seed germination, although micropropagation offers many advantages. Encapsulation technology has recently attracted the interest of researchers in the field of plant propagation because it combines the advantages of zygotic or gamic seeds with those of micropropagation. In this study, we examined the encapsulation of Carrizo citrange uninodal microcuttings (3–4 mm long) and evaluated the influence of the calcium alginate coating, a short time storage at cold temperature, and different sowing substrates on the viability and regrowth of the explants. A secondary aim was to develop an efficient protocol…
Use of seeds from local loquat ecotypes for rootstock production
2014
In Italy loquat (Eriobotrya japonica, Lindl) seedlings are commonly produced in nurseries as rootstocks for commercial loquat orchards. Seedling production is characterized by low germination percentage and a slow plant growth. The aim of this study was to evaluate local ecotypes seeds of loquat for seedlings production. Some ecotypes (Bianco dolce, BRT20, Claudia, Fiore, La Mantia and Marcenò) and international cultivars (Algerie, Golden Nugget, Magdal, Peluche and Tanaka) were compared. Loquat seeds of 11 different genotypes were collected from mature fruits in commercial orchards. Seeds were extracted and washed after separating from flesh. A pretreatment of 24 hr of seed soaking in a sol…
A study of thyme germplasm in Sicily
2009
hyme is characteristic of the Mediterranean and is of interest for its multifunctional nature. It is widely used in seasoning and flavouring, in herbal products and cosmetics. The versatility of this species means it is also useful in the protection of areas prone to erosion or an ornamental plant when designing low-maintenance gardens. Thyme germplasm constitutes a large part of the wild flora in Sicily and is a potentially useful source of genetic variability. On the basis of this, in the two-year period from 2004-2005, research was carried out on wild thyme accessions. These were used to evaluate both its productivity and individual biotypes with the functional characteristics most sough…
Yield and Fruit Quality Characterization of Eight Old Sicilian Apple Cultivars
2011
In this trial, we examined the quality of eight old Sicilian apple cultivars (‘Cannamelo’, ‘Cardinale’, ‘Gelata Cola’, ‘Gelata’, ‘Granatino’, ‘San Giuseppe’, ‘Virticchiaro’, ‘Zitella’) and a commercial one (‘Annurca’ traditional clone) through bio-agronomic performances and chemical-physical analyses. We also analyzed flowering time, ripening time, pomological characteristics, and generated a specific descriptor list that indicates a great variability among the cultivars. The data obtained showed interesting characteristics in ‘Granatino’, ‘Virticchiaro’, and ‘Zitella’, such as total soluble solid, fruit size, peel cover color, yield efficiency, and crop load.
